RESUMO

In Japan, the concentration of fluoride (F-) leached from rocks, such as tuff breccia, excavated in tunnel construction projects often exceeds the Japanese environmental standard of 0.8 mg/L. Because of this, proper disposal methods are necessary for managing F--bearing excavated rocks. One effective solution based on circular economy is the use of an adsorption layer system. This system can simultaneously prevent the migration of F- released from excavated rocks and allow the recycling of this construction waste material. To determine the most suitable material for the disposal of excavated F--bearing tuff breccia from a tunnel construction in Hokkaido, Japan, four types of natural geological materials (S-1, S-2, S-3, and S-4) obtained near the tunnel construction site, as well as three types of commercial adsorbents (calcium (Ca), magnesium (Mg), and CaMg adsorbents) were selected for evaluation. The batch adsorption test results showed that S-1 and S-4 had high adsorption capacities for F-, and the adsorption process followed the Langmuir isotherm. The adsorption of F- to the natural adsorbents was strongly influenced by the pH and the presence of bicarbonate ions (HCO3-), but unaffected by chloride (Cl-) and sulfate (SO42-). There was also a strong positive correlation between the abundance of amorphous aluminum (Al) and iron (Fe) extracted and the adsorption of F-, indicating the importance of ion exchange reactions associated with surface OH- in immobilizing F-. Meanwhile, the Mg-bearing adsorbent exhibited the highest adsorption affinity for F- among the commercial adsorbents. This was attributed to adsorption through electrostatic interactions and coprecipitation with magnesium hydroxide (Mg(OH)2) formed during the hydration of magnesium oxide (MgO). To effectively incorporate these adsorbents into the adsorption layer system, parameters such as permeability and residence time need to be determined in order to maximize the retention of F- through adsorption, ion exchange and coprecipitation reactions.

Ultrasound (US) triggered alterations in the viscoelastic behavior of the procaine-loaded ionically gelatinized pectin hydrogel matrix, and drug release was observed using a sono-device rheometer. The gel softened immediately upon activation of the ultrasound operated at 43 kHz and remained in a softened state throughout the irradiation. Upon cessation of ultrasound, the gel promptly reverted to its original hardness. This cycle of softening was consistently observed in ionically crosslinked pectin hydrogels, resulting in the promotion of procaine release, particularly with higher US power and lower calcium concentration. As the amount of loaded procaine increased, the gel weakened due to ion exchange with the calcium crosslinker and procaine. The most substantial release efficiency, reaching 82 % with a concentration of 32 µg/ml, was achieved when the hydrogels contained 0.03 % procaine within the gelatinized hydrogel medicine at a calcium concentration of 0.9 M, representing a six-fold increase compared to that without US. Notably, US exposure affected the 3D porous structure and degradation rate, leading to hydrogel collapse and facilitating medicine release. Additionally, the procaine-loaded pectin hydrogels with 0.9 M calcium exhibited improved fibroblast cell viability, indicating non-toxicity compared to those hydrogels prepared at a higher Ca2+ concentration of 2.4 M.

The small GTPases, Rab5 and Rab7, are key regulators at multiple stages of the endocytic/endolysosomal pathway, including fusion and maturation of endosomes. In yeast, Vps21p (Rab5 homolog) recruits a GEF for Rab7 and activates the downstream Ypt7p (Rab7 homolog) on endosomal membrane. Although the model of this sequential activation from Vps21p to Ypt7p in the endocytic pathway has been established, activation mechanism of Ypt7p in the Vps21p-independent pathway has not been completely clarified. Here we show that Ypt7p is activated and mediates vacuolar fusion in cells lacking all yeast Rab5 genes, VPS21, YPT52, and YPT53. We also demonstrate that deletion of both VPS21 and YPT7 genes cause severe defect in the AP-3 pathway as well as the CPY pathway although the AP-3 pathway is mostly intact in each vps21Δ or ypt7Δ mutant. Interestingly, in vps21Δ ypt7Δ mutant cargos trafficked via the VPS or endocytic pathway accumulate beside nucleus whereas cargo trafficked via the AP-3 pathway disperse in the cytosol. These findings suggest that Ypt7p is activated and plays a Rab5-independent role in the AP-3-mediated pathway.

RESUMO

The ammonia absorption process of sodium borohydride for ammonia storage has been studied by using the operando NMR and FT-IR measurements under various ammonia pressures. As a result, the characteristic variation in the chemical states of both materials due to the liquefaction has been clarified.
